The Study of Production and Maintenance of Germ-free Apodemus agrarius

Abstract
The present study was designed to develop domesticating Apodemus agrarius Pallas(Striped Field Mice: SFM), natural host of Hantaan virus as a new specific pathogen free animals (SPF) and evaluate microbiological efficiency test in terms of
microbiological monitoring. To develop A. agrarius as a SPF animal, domesticating SFM was performed using surgical vinyl isolator by hysterectomy. Hysterectomized Apodemus dam were reared in the vinyl isolator with the temperature keeping 23¡¾1¡É
and
humidity at 50-60% and air flow changing 15 times/h. Seventeen of pregnant SFM were hysterectomized and seven of sixteen derived dam were successfully weaned. Weaned SFM were moved in the Barrier Facility with cylinder for transport.
Microbiological
tests were performed in the SFM reared in the infection rearing draft system, hysterectomized dam in the vinyl isolator, and captured Apodemus. Infection of Hantaan virus was tested by IFA and RT-nested PCR with sera and lung, respectively. IFA
indicated that 3 groups sera turned out to be negative of antibody for Hantaan virus and RT0nested PCR turned out to be negative antigen for Hantaan virus in the SFM reared in infection rearing system and vinyl isolator. But captured SFM were
possessed
antigen for Hantaan virus. RT-PCR for HVJ indicated that lung tissue of 3 group turned out to be negative antigen. Also RT-PCR for MHV indicated that liver tissue of 3 group turned out to be negative. And only E. coli was identified in the small
intestine and cecum of hysterectomized dam and any other parasites were not found. It is concluded that successful derivation of Apodemus dam to the barrier facility, we are able to produce SFM as a SPF status and subsequently provide SFM to
various
researches such as microbiology, agricultural medicine, etc.
